Well this would require a great deal of everything really. You would need a Flash or HTML5 knowledge to play the video. SQL databases to keep everything from play count to account details in order. PHP because you’ll need dynamic pages. On the server you’ll need to be able to convert video that people upload to the format you’ll be streaming. Streaming video takes up a lot of resources and most hosters either won’t allow it or require you to pay a lot more because you are taking up a lot of CPU power and Bandwidth on a single server.
This is definitely not the project for a beginner, especially since it is already been done so many times.
